目的:研究环孢霉素A( Cyclosporin A, CsA )对不同情况下的兔晶状体上皮细胞(rabbit lens epithelial cells, RLECs)超微结构和增殖的影响。方法:用CsA分别处理正常生长的RLECs和经过氧化氢(hydrogen peroxide, H2O2)损伤的RLECs后,用透射电镜观察RLECs超微结构变化;MTT法测定RLECs增殖能力改变。结果:CsA浓度≥10µmol•L-1时,明显损伤正常RLECs的超微结构和增殖;当CsA浓度<40µmol•L-1时,能部分抑制H2O2对RLECs超微结构损伤和增殖抑制,对细胞的具有保护作用,其中以10µmol•L-1的CsA对RLECs的保护作用最强,与正常对照组比较,差异无显著意义(P=0.4013)。结论:环孢霉素A对不同情况下的兔晶状体上皮细胞的超微结构和增殖具有不同的作用。
【Abstract】Objective To investigate the effect of cyclosporine A on the ultrastructure and proliferation of lens epithelial cells. Methods Lens epithelial cells in different conditions were treated with cyclosporine A. Using transmission electron microscope and MTT assay, we observed the changes of ultrastructure and proliferation of the lens epithelial cells respectively. Results Cyclosporine A beyond 10µmol•L-1 destroy the ultrastructure and proliferation of lens epithelial cells of rabbit; Cyclosporine A less than 40µmol•L-1 can partially protect the lens epithelial cells’ ultrastructure and proliferation from hydrogen peroxide. Simultaneously, there was a little difference between the results of 10µmol•L-1 Cyclosporine A protected group and control group(P=0.4013). Conclusion cyclosporine A has opposite effects on rabbit lens epithelial cells in different conditions.
