目的 检测不同剂量微波辐射对兔晶状体上皮细胞(rabbit lens epithelial cells, RLECs)细胞周期的阻断作用,并探讨微波辐射对细胞周期调控基因P21WAP1、P27Kip1和c-myc表达的影响。方法 应用频率2450MHz,功率密度分别为0.10mW/cm2、0.25 mW/cm2、0.50 mW/cm2、1.00 mW/cm2、2.00 mW/cm2的微波连续辐照体外培养的RLECs 8h,光镜下观察辐射前后细胞形态学改变,采用流式细胞技术测定细胞周期,Western印迹法检测2.00 mW/cm2微波辐照 4h 、6h和8h后对P21WAP1、P27Kip1和c-myc基因表达的影响。结果 高于0.50 mW/cm2的微波辐射8h后,即可引起RLECs肿胀和聚集,甚至出现细胞固缩和脱壁现象,同时使G0/G1期细胞所占比例明显升高, 0.10mW/cm2和0.25 mW/cm2辐射组与对照组比较未见明显变化。2.00mW/cm2微波辐照后P27Kip1表达水平明显升高而c-myc表达水平下降, P21WAP1表达在辐射前后无变化。结论 功率密度高于0.50 mW/cm2的微波即可引起体外培养的晶体上皮细胞产生形态学改变,并阻断细胞周期使之停滞于G0/G1期,这种阻断作用可能通过分别调节P27Kip1和c-myc这两个调控细胞周期转换的基因而实现的,对P21WAP1的表达不影响。

[Abstract] Objective To study the cell cycle block induced by different doses of microwave in cultured rabbit lens epithelial cells (RLECs) and its effects on the gene expression of P21WAP1, P27Kip1 and c-myc. Methods Cultured rabbit lens epithelial cells were exposed to microwave radiation with frequency of 2450MHz and power density of 0.10 mW/cm2, 0.25 mW/cm2, 0.50 mW/cm2, 1.00 mW/cm2 and 2.00 mW/cm2 for 8 hours in vitro. The morphological change of RLECs was observed under microscop, effects of microwave radiation on RLECs cell cycle were measured by flow cytometry. Expression of P21WAP1, P27Kip1 and c-myc were estimated by western blot after 2.00mW/cm2 microwave radiation for 4 hours, 6 hours and 8 hours. Results 8 hours after radiation, RLECs treated by 0.50mW/cm2, 1.00mW/cm2 and 2.00mW/cm2 microwave showed swelling, congregation and detachment. The flow cytometric analysis demonstrated that microwave blocked cell cycle at G0/G1 phase. No obvious change could be detected in 0.10 mW/cm2 and 0.25mW/cm2 microwave groups. The gene expression of P27Kip1 was increased and c-myc was decreased after 2.00 mW/cm2 radiation, it did not affect P21WAP1. Conclusion Microwave exceeding 0.50mW/cm2 can block RLECs cell cycle at G0/G1 phase, which may be related to regulation of P27Kip1 and c-myc expression, but not P21WAP1.
