目的 观察中药半边旗二萜类化合物5F对体外培养的人翼状胬肉成纤维细胞(HPFs)凋亡的影响以及对Fas基因表达的调控,初步探讨该药对HPF作用的机制。方法 应用荧光染色观察HPF形态改变,流式细胞术(FCM)分析不同浓度药物对HPF周期分布及凋亡的诱导,免疫细胞化学法(ICH)检测5F对凋亡调控基因Fas表达的影响。结果 5F浓度32μg/ml作用不同时间诱导HPF凋亡。5F使Fas基因表达上调。8μg/ml浓度时Fas阳性表达率与对照组比较无显著性差异(P>0.05),药量增至32μg/ml后Fas阳性表达率增加,与对照组比较差异显著,各浓度组间丙两两比较亦有显著差异(P<0.01)。结论 在一定浓度下5F诱导HPF凋亡,随用药时间延长细胞由凋亡发展为坏死,5F对HPF增殖的抑制作用与促凋亡基因Fas表达上调密切相关,提示Fas表达可作为检测HPF抑制的主要指标,5F具潜在临床价值。

[Abstract] Objective To observe the effect of diterpenoid compound 5F of Pteris semipinnate L on apoptosis, and expressions of Fas of cultured human pterygium body fibroblasts (HPF),thereby evaluating the mechanism of action of this agent. Methods The morphologic changes of HPFs were observed microscopically with fluorescent stain. The changes of the cell cycle and apoptotic induction of HPFs were evaluated with flow cytometry (FCM). The expressions of Fas were determined by SP method. Results 5F at the concentration of 32μg/ml induced HPF apoptosis at various time points. The expression of Fas on HPFs treated with 5F at the concentration of 8μg/ml was similar to that in control group (P>0.05);they were added to 24.9% at 32μg/ml of the drug. There existed significant differences in the expression between two groups and intra-cla sses (P<0.01). Conclusion 5F could induce HPF apoptosis at a special strength, and apoptotic cells were evolved to necrosis with the prolongation of the time. The inhibitory effect of 5F on HPFs may be related to up-regulation of expression of Fas. It is suggested that the expressions of Fas could serve as major markings for the detection of HPF proliferation or inhibition.5F has a potential clinical value.
