Objective: To test the feasibility of recording rod ON and OFF response in electroretinograms and multifocal ERG, and investigate appropriate recording conditions for clinical use. Methods: Rod On and OFF response were recorded using a rapid-on and rapid -off (sawtooth) stimulus in humans after dark adaptation. Multifocal ERGs flashes were blue (W47B), blank frames between two successive flashes of one hexagon was varied from 0 to 8. Flash frequency ,flash intensities in ERG were varied to obtain appropriate recording conditions for clinical use. Results: Rod On and OFF response were clearly recordable.The waveform and timing of ON response were similar to full-field rod ERGs, OFF response was negtive wave. With increasing stimulus intensity, the On response incerased,while OFF response change little. Increasing stimulus frequency, the amplitude of ON and OFF response decreased. Conclusions: Rod ON and OFF response can be recorded in human . The stimulus frequency of 1 Hz in ERG were suggested to appropriate recording conditions for clinical use.
